Minimum and Optimal DNA Requirements (The client has the option to provide us starting material, Intact Genomics will prepare DNA with additional cost for sequencing):

gDNA:

  • Provide 1-3ug of high quality, high molecular weight gDNA in water or 10 mM Tris-HCL pH 8.0.
  • Please quantify DNA by fluorometric methods if possible.  If quantifying using a spectrophotometer, A260/A280 should be around 1.8.  We also recommend to send more DNA for QC purposes.
  • Please provide a gel image to assess DNA quality – a good image will have no smearing and a distinct band above 10kb.
  • We strongly recommend column based cleanups or ethanol precipitation as well, especially for DNA prepared by organic extraction.

RNA:

  • Provide 1 ug of total RNA or 100ng-1ug of mRNA in water or 10 mM Tris-HCL pH 8.
  •  A260/A280 ratio should be in the range of 1.8 to 2.1.
  • Please ribosomally reduce Prokaryotic total RNA prior to sample submission.
  • We strongly recommend column based cleanup, especially for organic extractions. We also recommend assaying ribosomal peak integrity on an Agilent Bioanalyzer PicoChip. 

small RNA:

  • Provide 1 ug of total RNA in water or 10 mM Tris-HCL pH 8.0.
  • A260/A280 ratios should be in the range of 1.8 to 2.1.
  • We strongly recommend column based cleanup, especially for organic extractions. We also recommend assaying ribosomal peak integrity on an Agilent Bioanalyzer PicoChip.

ChIP:

  • Provide at least 10 ng, 30 ng would be optimal.   Post immunoprecipitated DNA should be sheared to 200-600 bp.
  • Please don’t share DNA to >1000 bp.
  • Please quantify DNA by fluorometric methods if possible.  If quantifying using aspectrophotometer, A260/A280 should be between 1.7 and 1.9.  We also recommend sending 3x as much DNA (30-90 ng) for QC purposes.
  • We also recommend performing qPCR on a known target to verify enrichment
  • Also, please submit an input sample as a sequencing control.

Shipping / Pick-up:

If you are submitting more than 12 samples, please provide samples to Intact Genomics in full-skirted 96-well plates.  The 96-well plates should be fully sealed utilizing either foil plate seals or caps.  Using a permanent marker, legibly label plates with Unique plate name, Customer last name and Date.

If you are submitting fewer than 12 samples, provide samples in either a 0. 5 ml or 1.5 ml nuclease-free micro centrifuge tube.  Include the sample name clearly on the cap using no more than 8 letters or numbers.  Please seal samples in a plastic bag.

Also please provide a spreadsheet with sample information accompanies the plate/tube, and emailed the spreadsheet to: sales@intactgenomics.com.
